What are the methylation reactions of 2-thiouracil?
2-thiouracil (12.8 g) and fine powdered sodium hydroxide (4.3 g) in water (35 mL) in a dry reaction flask, and then heat the resulting reaction mixture to 60-70 degree C until the solid dissolves. Then add ethanol (70 mL) to the mixture and cool the resulting reaction solution to around 30 degree C. Add 6.3 mL of iodomethane to the mixture. Heat the solution again to 50-60 degree C and keep it in that environment while stirring for about 20 minutes. Monitor the reaction progress through TLC spot plates. After the raw materials in the reaction system have completely reacted, cool the reaction mixture to room temperature, and then filter and collect the precipitated white solid. The obtained reaction mixture can be purified by recrystallization from ethanol to obtain the methylated target product molecule.
